Specificity    This assay has high sensitivity and excellent specificity for detection of C8alpha. No significant cross-reactivity or interference between C8alpha and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between C8alpha and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of C8A elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for C8A purchase    MBS7228843 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Complement component C8 alpha chain (C8A) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing C8A, with an estimated sensitivity of 0.1 ng/mL. The ELISA analytical biochemical technique of the MBS7228843 kit is based on C8A antibody-C8A antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect C8A antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, C8A. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for C8A elisa kit

Intended Uses: This C8alpha ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse C8alpha. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay||C8alpha ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-C8alpha antibody and an C8alpha-HRP conjugate. The assay sample and buffer are incubated together with C8alpha-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the C8alpha concentration since C8alpha from samples and C8alpha-HRP conjugate compete for the anti-C8alpha antibody binding site. Since the number of sites is limited, as more sites are occupied by C8alpha from the sample, fewer sites are left to bind C8alpha-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The C8alpha concentration in each sample is interpolated from this standard curve.

NCBI Summary for C8A    C8 is a component of the complement system and contains three polypeptides, alpha, beta and gamma. This gene encodes the alpha subunit of C8. C8 participates in the formation of the membrane attack complex (MAC). The MAC assembles on bacterial membranes to form a pore, permitting disruption of bacterial membrane organization. Mutations in this gene cause complement C8 alpha-gamma deficiency. [provided by RefSeq, Nov 2008]
Section Bar
UniProt Comments for C8A    C8A: Constituent of the membrane attack complex (MAC) that plays a key role in the innate and adaptive immune response by forming pores in the plasma membrane of target cells. C8A inserts into the target membrane, but does not form pores by itself. Defects in C8A are a cause of complement component 8 deficiency type 1 (C8D1). A rare defect of the complement classical pathway associated with susceptibility to severe recurrent infections, predominantly by Neisseria gonorrhoeae or Neisseria meningitidis. Belongs to the complement C6/C7/C8/C9 family.

Protein type: Membrane protein, multi-pass

Chromosomal Location of Human Ortholog: 1p32

Research Articles on C8A    1. C8 alpha and C8 beta have correspondingly similar roles in MAC-mediated lysis of erythrocytes and bacterial killing. C8 gamma is not required for complement-mediated killing of Gram-negative bacteria.
