 Assay Procedure Summary    1. Prepare all reagents, samples and standards;
2. Add 100uL standard or sample to each well. Incubate 2 hours at 37 degree C;
3. Aspirate and add 100uL prepared Detection Reagent A. Incubate 1 hour at 37 degree C;
4. Aspirate and wash 3 times;
5. Add 100uL prepared Detection Reagent B. Incubate 30 minutes at 37 degree C;
6. Aspirate and wash 5 times;
7. Add 90uL Substrate Solution. Incubate 15-25 minutes at 37 degree C;
8. Add 50uL Stop Solution. Read at 450nm immediately.

 Preparation and Storage    For unopened kit: All the reagents should be kept according to the labels on vials. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C upon receipt while the others should be at 4 degree C.

For opened kit: When the kit is opened, the remaining reagents still need to be stored according to the above storage condition. Besides, please return the unused wells to the foil pouch containing the desiccant pack, and reseal along entire edge of zip-seal.

The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

Product Description specifically for CRYbB2 elisa kit

   Principle of the Assay: The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Crystallin Beta B2 (CRYbB2). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Crystallin Beta B2 (CRYbB2). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Crystallin Beta B2 (CRYbB2), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Crystallin Beta B2 (CRYbB2) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

 NCBI Summary for CRYbB2    Crystallins are separated into two classes: taxon-specific, or enzyme, and ubiquitous. The latter class constitutes the major proteins of vertebrate eye lens and maintains the transparency and refractive index of the lens. Since lens central fiber cells lose their nuclei during development, these crystallins are made and then retained throughout life, making them extremely stable proteins. Mammalian lens crystallins are divided into alpha, beta, and gamma families; beta and gamma crystallins are also considered as a superfamily. Alpha and beta families are further divided into acidic and basic groups. Seven protein regions exist in crystallins: four homologous motifs, a connecting peptide, and N- and C-terminal extensions. Beta-crystallins, the most heterogeneous, differ by the presence of the C-terminal extension (present in the basic group, none in the acidic group). Beta-crystallins form aggregates of different sizes and are able to self-associate to form dimers or to form heterodimers with other beta-crystallins. This gene, a beta basic group member, is part of a gene cluster with beta-A4, beta-B1, and beta-B3. A chain-terminating mutation was found to cause type 2 cerulean cataracts.
