Specificity    This assay has high sensitivity and excellent specificity for detection of NPC2. No significant cross-reactivity or interference between NPC2 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between NPC2 and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of NPC2 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for NPC2 purchase    MBS7206139 is a ready-to-use microwell, strip plate Competitive (Quantitative) ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Epididymal secretory protein E1 (NPC2) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing NPC2, with an estimated sensitivity of 0.1 ng/mL. The ELISA analytical biochemical technique of the MBS7206139 kit is based on NPC2 antibody-NPC2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect NPC2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, NPC2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for NPC2 elisa kit

Intended Uses: This NPC2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse NPC2. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: NPC2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-NPC2 antibody and an NPC2-HRP conjugate. The assay sample and buffer are incubated together with NPC2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NPC2 concentration since NPC2 from samples and NPC2-HRP conjugate compete for the anti-NPC2 antibody binding site. Since the number of sites is limited, as more sites are occupied by NPC2 from the sample, fewer sites are left to bind NPC2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NPC2 concentration in each sample is interpolated from this standard curve.

NCBI Summary for NPC2    This gene encodes a protein containing a lipid recognition domain. The encoded protein may function in regulating the transport of cholesterol through the late endosomal/lysosomal system. Mutations in this gene have been associated with Niemann-Pick disease, type C2 and frontal lobe atrophy. [provided by RefSeq, Jul 2008]

UniProt Comments for NPC2    NPC2: Intracellular cholesterol transporter which acts in concert with NPC1 and plays an important role in the egress of cholesterol from the endosomal/lysosomal compartment. Both NPC1 and NPC2 function as the cellular 'tag team duo' (TTD) to catalyze the mobilization of cholesterol within the multivesicular environment of the late endosome (LE) to effect egress through the limiting bilayer of the LE. NPC2 binds unesterified cholesterol that has been released from LDLs in the lumen of the late endosomes/lysosomes and transfers it to the cholesterol-binding pocket of the N-terminal domain of NPC1. Cholesterol binds to NPC1 with the hydroxyl group buried in the binding pocket and is exported from the limiting membrane of late endosomes/ lysosomes to the ER and plasma membrane by an unknown mechanism. The secreted form of NCP2 regulates biliary cholesterol secretion via stimulation of ABCG5/ABCG8-mediated cholesterol transport. Defects in NPC2 are the cause of Niemann-Pick disease type C2 (NPDC2). A lysosomal storage disorder that affects the viscera and the central nervous system. It is due to defective intracellular processing and transport of low-density lipoprotein derived cholesterol. It causes accumulation of cholesterol in lysosomes, with delayed induction of cholesterol homeostatic reactions. Niemann-Pick disease type C2 has a highly variable clinical phenotype. Clinical features include variable hepatosplenomegaly and severe progressive neurological dysfunction such as ataxia, dystonia and dementia. The age of onset can vary from infancy to late adulthood. Belongs to the NPC2 family.
