Preparation and Storage    The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of KARS elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for KARS purchase    MBS2023685 is a ready-to-use microwell, strip plate Double-antibody Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Lysyl tRNA Synthetase (KARS) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 31.2-2000pg/mL in biological research samples containing KARS, with an estimated sensitivity of < 12.9pg/mL. The ELISA analytical biochemical technique of the MBS2023685 kit is based on KARS antibody-KARS antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect KARS antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, KARS. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, Plasma, Tissue homogenates and Other Biological Fluids. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for KARS elisa kit

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Lysyl tRNA Synthetase (KARS). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Lysyl tRNA Synthetase (KARS). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Lysyl tRNA Synthetase (KARS), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Lysyl tRNA Synthetase (KARS) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Summary for KARS    Aminoacyl-tRNA synthetases are a class of enzymes that charge tRNAs with their cognate amino acids. Lysyl-tRNA synthetase is a homodimer localized to the cytoplasm which belongs to the class II family of tRNA synthetases. It has been shown to be a target of autoantibodies in the human autoimmune diseases, polymyositis or dermatomyositis. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

UniProt Comments for KARS    KARS: Catalyzes the specific attachment of an amino acid to its cognate tRNA in a 2 step reaction: the amino acid (AA) is first activated by ATP to form AA-AMP and then transferred to the acceptor end of the tRNA. When secreted, acts as a signaling molecule that induces immune response through the activation of monocyte/macrophages. Catalyzes the synthesis of diadenosine oligophosphate (Ap4A), a signaling molecule involved in the activation of MITF transcriptional activity. Interacts with HIV-1 virus GAG protein, facilitating the selective packaging of tRNA(3)(Lys), the primer for reverse transcription initiation. Homodimer; also part of a multisubunit complex that groups AIMP1, AIMP2, EEF1A1 and tRNA ligases for Arg, Asp, Glu, Gln, Ile, Leu, Lys, Met and Pro. Interacts with AIMP2 (via N- terminus) and MITF. Interacts directly with HIV-1 virus GAG protein. Up-regulated by DARS and EEF1A1, but not by AIMP2. Belongs to the class-II aminoacyl-tRNA synthetase family. 2 isoforms of the human protein are produced by alternative splicing.
