The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 0.625-40ng/mL in biological research samples containing MBP, with an estimated sensitivity of <0.249ng/mL. The ELISA analytical biochemical technique of the MBS2022666 kit is based on MBP antibody-MBP antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MBP antigen targets in samples.    Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of MBP in mouse serum, plasma, tissue homogenates, cell lysates and other biological fluids.

Principle of the Assay: The microplate provided in this kit has been pre-coated with an antibody specific to MBP. Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to MBP. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain MBP, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of MBP in the samples is then determined by comparing the O.D. of the samples to the standard curve.

 NCBI Summary for MBP    The protein encoded by this gene is the predominant constituent of the crystalline core of the eosinophil granule. High levels of the proform of this protein are also present in placenta and pregnancy serum, where it exists as a complex with several other proteins including pregnancy-associated plasma protein A (PAPPA), angiotensinogen (AGT), and C3dg. This protein may be involved in antiparasitic defense mechanisms as a cytotoxin and helminthotoxin, and in immune hypersensitivity reactions. The encoded protein contains a peptide that displays potent antimicrobial activity against Gram-positive bacteria, Gram-negative bacteria, and fungi. It is directly implicated in epithelial cell damage, exfoliation, and bronchospasm in allergic diseases.
