The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 0.312-20 ng/mL in biological research samples containing Sirt3, with an estimated sensitivity of 0.156 ng/mL. The ELISA analytical biochemical technique of the MBS2885911 kit is based on Sirt3 antibody-Sirt3 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect Sirt3 antigen targets in samples.    Intended Uses: This immunoassay kit allows for the in vitro quantitative determination of target antigen concentrations in serum, plasma, tissue homogenates, cell culture supernates or other biological fluids.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to target antigen. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for target antigen and then avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain target antigen, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of target antigen in the samples is then determined by comparing the O.D. of the samples to the standard curve.

 UniProt Comments for Sirt3    SIRT3: a soluble mitochondrial NAD-dependent protein deacetylase. Can deacetylate and thereby activate a central metabolic regulator in the mitochondrial matrix, glutamate dehydrogenase. Mitochondrial proteins are hyperacetylated in SIRT3-deficient mice, but not in SIRT4 or SIRT5 deficient mice. SIRT3-deficient mice have no obvious metabolic phenotype, and the process of adaptive thermogenesis is normal. Can deacetylate and activate isocitrate dehydrogenase 2, an enzyme that participates in regulation of the citric acid cycle and the regeneration of antioxidants. Inhibited by nicotinamide, but not by trichostatin A. Its unprocessed form is enzymatically inactive. It is processed by mitochondrial processing peptidase (MPP) in the mitochondrial matrix to give an enzymatically active 28 kDa product.
