Other Notes    Small volumes of PRF1 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for PRF1 purchase    MBS263793 is a ready-to-use microwell, strip plate Quantitative Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Perforin (PRF1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing PRF1, with an estimated sensitivity of Up to 0.5 ng/ml. The ELISA analytical biochemical technique of the MBS263793 kit is based on PRF1 antibody-PRF1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PRF1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PRF1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Mouse serum, plasma or cell culture supernatant and organizations in the natural and recombinant PRF concentration. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for PRF1 elisa kit

Principle of the Assay: This experiment use double-sandwich elisa technique and the ELISA Kit provided is typical. The pre-coated antibody is Mouse PRF monoclonal antibody and the detecting antibody is polyclonal antibody with biotin labeled. Samples and biotin labeling antibody are added into ELISA plate wells and washed out with PBS or TBS. Then Avidin-peroxidase conjugates are added to ELISA wells in order; Use TMB substrate for coloring after reactant thoroughly washed out by PBS or TBS. TMB turns into blue in peroxidase catalytic and finally turns into yellow under the action of acid. The color depth and the testing factors in samples are positively correlated.

NCBI Summary for PRF1    The protein encoded by this gene has structural and functional similarities to complement component 9 (C9). Like C9, this protein creates transmembrane tubules and is capable of lysing non-specifically a variety of target cells. This protein is one of the main cytolytic proteins of cytolytic granules, and it is known to be a key effector molecule for T-cell- and natural killer-cell-mediated cytolysis. Defects in this gene cause familial hemophagocytic lymphohistiocytosis type 2 (HPLH2), a rare and lethal autosomal recessive disorder of early childhood. Alternative splicing results in multiple transcript variants encoding the same protein. [provided by RefSeq, Jul 2008]

UniProt Comments for PRF1    PRF1: Plays a key role in secretory granule-dependent cell death, and in defense against virus-infected or neoplastic cells. Plays an important role in killing other cells that are recognized as non-self by the immune system, e.g. in transplant rejection or some forms of autoimmune disease. Can insert into the membrane of target cells in its calcium-bound form, oligomerize and form large pores. Promotes cytolysis and apoptosis of target cells by facilitating the uptake of cytotoxic granzymes. Monomer, as sobluble protein. Homooligomer. Oligomerization is required for pore formation. Repressed by contact with target cells. Belongs to the complement C6/C7/C8/C9 family.

Protein type: Membrane protein, multi-pass

Chromosomal Location of Human Ortholog: 10q22

Cellular Component: membrane; cytoplasmic membrane-bound vesicle; plasma membrane; integral to membrane; extracellular region

Molecular Function: protein binding; calcium ion binding; wide pore channel activity

Biological Process: formation of immunological synapse; apoptosis; cytolysis; defense response to tumor cell; cellular defense response; immune response to tumor cell; defense response to virus; transmembrane transport; protein homooligomerization

Disease: Lymphoma, Non-hodgkin, Familial; Aplastic Anemia; Hemophagocytic Lymphohistiocytosis, Familial, 2

Research Articles on PRF1    1. These data show that monoallelic germline mutations of PRF1 are frequent in patients with childhood anaplastic large cell lymphoma, suggesting that partially impaired cytotoxic machinery may represent a predisposing factor for it.
