Specificity    This assay has high sensitivity and excellent specificity for detection of PSA. No significant cross-reactivity or interference between PSA and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between PSA and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of PSA elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for PSA purchase    MBS729544 is a ready-to-use microwell, strip plate Quantitative Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Prostate specific antigen (PSA) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing PSA, with an estimated sensitivity of 1.0 pg/mL. The ELISA analytical biochemical technique of the MBS729544 kit is based on PSA antibody-PSA antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PSA antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PSA. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for PSA elisa kit

Intended Uses: This PSA ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse PSA. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: PSA ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for PSA. Standards or samples are then added to the microtiter plate wells and PSA if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of PSA present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for PSA are added to each well to "sandwich" the PSA immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain PSA and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PSA concentration in each sample is interpolated from this standard curve.

NCBI Summary for PSA    Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. Its protein product is a protease present in seminal plasma. It is thought to function normally in the liquefaction of seminal coagulum, presumably by hydrolysis of the high molecular mass seminal vesicle protein. Serum level of this protein, called PSA in the clinical setting, is useful in the diagnosis and monitoring of prostatic carcinoma. Alternate splicing of this gene generates several transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]

UniProt Comments for PSA    PSA: Hydrolyzes semenogelin-1 thus leading to the liquefaction of the seminal coagulum. Forms heterodimer with SERPINA5. Inhibited by SERPINA5. Activity is strongly inhibited by Zn2+, 100 times more abundant in semen than in serum. This inhibition is relieved by exposure to semenogelins, which are avid zinc binders. Belongs to the peptidase S1 family. Kallikrein subfamily.
