Principle of the assay: SEPP-1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SEPP-1 antibody and an SEPP-1-HRP conjugate. The assay sample and buffer are incubated together with SEPP-1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SEPP-1 concentration since SEPP-1 from samples and SEPP-1-HRP conjugate compete for the anti-SEPP-1 antibody binding site. Since the number of sites is limited, as more sites are occupied by SEPP-1 from the sample, fewer sites are left to bind SEPP-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SEPP-1 concentration in each sample is interpolated from this standard curve.

 NCBI Summary for SEPP1    This gene encodes a selenoprotein containing multiple selenocysteine (Sec) residues encoded by the UGA codon that normally signals translation termination. The 3' UTR of selenoprotein genes have a common stem-loop structure, the sec insertion sequence (SECIS), that is necessary for the recognition of UGA as a Sec codon rather than as a stop signal. This selenoprotein is a secreted protein, and is unusual in that it contains 10 selenocysteine residues per polypeptide, constituting most of the selenium in plasma. It has been implicated as an extracellular antioxidant and in the transport of selenium. Transcript variants differing in the 5' UTR, but encoding the same protein, have been found for this gene.
