Specificity    This assay has high sensitivity and excellent specificity for detection of TSPO. No significant cross-reactivity or interference between TSPO and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between TSPO and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of TSPO elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for TSPO purchase    MBS724094 is a ready-to-use microwell, strip plate Competitive or Sandwich (Quantitative) ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Translocator protein (TSPO) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing TSPO, with an estimated sensitivity of 0.1 ng/mL. The ELISA analytical biochemical technique of the MBS724094 kit is based on TSPO antibody-TSPO antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TSPO antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TSPO. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for TSPO elisa kit

Intended Uses: This TSPO ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse TSPO. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: TSPO ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-TSPO antibody and an TSPO-HRP conjugate. The assay sample and buffer are incubated together with TSPO-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TSPO concentration since TSPO from samples and TSPO-HRP conjugate compete for the anti-TSPO antibody binding site. Since the number of sites is limited, as more sites are occupied by TSPO from the sample, fewer sites are left to bind TSPO-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TSPO concentration in each sample is interpolated from this standard curve.

NCBI Summary for TSPO    Present mainly in the mitochondrial compartment of peripheral tissues, the protein encoded by this gene interacts with some benzodiazepines and has different affinities than its endogenous counterpart. The protein is a key factor in the flow of cholesterol into mitochondria to permit the initiation of steroid hormone synthesis. Alternatively spliced transcript variants have been reported; one of the variants lacks an internal exon and is considered non-coding, and the other variants encode the same protein. [provided by RefSeq, Feb 2012]

UniProt Comments for TSPO    TSPO: Responsible for the manifestation of peripheral-type benzodiazepine recognition sites and is most likely to comprise binding domains for benzodiazepines and isoquinoline carboxamides. May play a role in the transport of porphyrins and heme. Plays a role in the transport of cholesterol across mitochondrial membranes in steroidogenic cells. Belongs to the TspO/BZRP family. Interacts with BZRAP1. May interact with STAR. Interacts with MOST-1. 2 isoforms of the human protein are produced by alternative splicing.
