Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact technical support.
Other Notes    Small volumes of PLAUR/Upar elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for PLAUR/Upar purchase    MBS726020 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Urokinase-type Plasminogen Activator Receptor (PLAUR/Upar) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards render a theoretical kit detection range in biological research samples containing PLAUR/Upar, with an estimated sensitivity of 1.0 pg/mL. The ELISA analytical biochemical technique of the MBS726020 kit is based on PLAUR/Upar antibody-PLAUR/Upar antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PLAUR/Upar antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PLAUR/Upar. Appropriate sample types may include undiluted human body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for PLAUR/Upar elisa kit

Principle of the Assay: uPAR ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for uPAR Standards or samples are then added to the microtiter plate wells and uPAR if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of uPAR present in the sample, a standardized preparation of horseradish peroxidase (HRP) -conjugated polyclonal antibody specific for uPAR are added to each well to "sandwich" the uPAR immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. Hie enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain uPAR and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotonietrically at a wavelength of 450 nm A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The uPAR concentration in each sample is interpolated from this standard curve.

NCBI Summary for PLAUR/Upar    This gene encodes the receptor for urokinase plasminogen activator and, given its role in localizing and promoting plasmin formation, likely influences many normal and pathological processes related to cell-surface plasminogen activation and localized degradation of the extracellular matrix. It binds both the proprotein and mature forms of urokinase plasminogen activator and permits the activation of the receptor-bound pro-enzyme by plasmin. The protein lacks transmembrane or cytoplasmic domains and may be anchored to the plasma membrane by a glycosyl-phosphatidylinositol (GPI) moiety following cleavage of the nascent polypeptide near its carboxy-terminus. However, a soluble protein is also produced in some cell types. Alternative splicing results in multiple transcript variants encoding different isoforms. The proprotein experiences several post-translational cleavage reactions that have not yet been fully defined. [provided by RefSeq, Jul 2008]

UniProt Comments for PLAUR/Upar    uPAR: Acts as a receptor for urokinase plasminogen activator. Plays a role in localizing and promoting plasmin formation. Mediates the proteolysis-independent signal transduction activation effects of U-PA. It is subject to negative-feedback regulation by U-PA which cleaves it into an inactive form. Monomer (Probable). Interacts with MRC2. Interacts (via the UPAR/Ly6 domains) with SRPX2. Expressed in neurons of the rolandic area of the brain. Expressed in the brain. 3 isoforms of the human protein are produced by alternative splicing.
