UniProt Comments for ALKBH3
ALKBH3: Dioxygenase that repairs alkylated DNA containing 1- methyladenine (1meA) and 3-methylcytosine (3meC) by oxidative demethylation. Has a strong preference for single-stranded DNA. Able to process alkylated 3mC within double-stranded regions via its interaction with ASCC3, which promotes DNA unwinding to generate single-stranded substrate needed for ALKHB3. May also act on RNA. Requires molecular oxygen, alpha-ketoglutarate and iron. Belongs to the alkB family. 2 isoforms of the human protein are produced by alternative splicing.
Protein type: Oxidoreductase; Hydrolase; Demethylase; DNA repair, damage; EC 1.14.11.-
Cellular Component: nucleoplasm; mitochondrion; cytoplasm; nucleus
Molecular Function: procollagen-proline dioxygenase activity; 2,4-dichlorophenoxyacetate alpha-ketoglutarate dioxygenase activity; sulfonate dioxygenase activity; dioxygenase activity; ferrous iron binding; endonuclease activity; metal ion binding; exonuclease activity; DNA-N1-methyladenine dioxygenase activity; oxidoreductase activity, acting on paired donors, with incorporation or reduction of molecular oxygen, 2-oxoglutarate as one donor, and incorporation of one atom each of oxygen into both donors; oxidoreductase activity; DNA demethylase activity
Biological Process: cell proliferation; DNA dealkylation; DNA repair; response to DNA damage stimulus
