The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 3.125 pg/ml-200 pg/ml in biological research samples containing ANGPTL4, with an estimated sensitivity of 0.781 pg/ml. The ELISA analytical biochemical technique of the MBS915491 kit is based on ANGPTL4 antibody-ANGPTL4 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ANGPTL4 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ANGPTL4. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, tissue homogenates. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with ANGPTL4. Standards or samples are added to the appropriate microtiter plate wells with Horseradish Peroxidase (HRP) conjugated antibody preparation specific for ANGPTL4. The competitive inhibition reaction is launched between with pre-coated ANGPTL4 and ANGPTL4 in samples. A substrate solution is added to the wells and the color develops in opposite to the amount of ANGPTL4 in the samples. The color development is stopped and the intensity of the color is measured.

ANGPTL4: Protein with hypoxia-induced expression in endothelial cells. May act as a regulator of angiogenesis and modulate tumorigenesis. Inhibits proliferation, migration, and tubule formation of endothelial cells and reduces vascular leakage. May exert a protective function on endothelial cells through an endocrine action. It is directly involved in regulating glucose homeostasis, lipid metabolism, and insulin sensitivity. In response to hypoxia, the unprocessed form of the protein accumulates in the subendothelial extracellular matrix (ECM). The matrix-associated and immobilized unprocessed form limits the formation of actin stress fibers and focal contacts in the adhering endothelial cells and inhibits their adhesion. It also decreases motility of endothelial cells and inhibits the sprouting and tube formation.
