Specificity    This assay has high sensitivity and excellent specificity for detection of ARR. No significant cross-reactivity or interference between ARRand analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between ARRand all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of COL4A1 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for COL4A1 purchase    MBS7202946 is a ready-to-use microwell, strip plate Quantitative Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the arresten (COL4A1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing COL4A1, with an estimated sensitivity of 1.0 ng/mL. The ELISA analytical biochemical technique of the MBS7202946 kit is based on COL4A1 antibody-COL4A1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect COL4A1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, COL4A1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This ARRELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse ARR. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: ARRELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ARRantibody and an ARR-HRP conjugate. The assay sample and buffer are incubated together with ARR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ARRconcentration since ARRfrom samples and ARR-HRP conjugate compete for the anti-ARRantibody binding site. Since the number of sites is limited, as more sites are occupied by ARRfrom the sample, fewer sites are left to bind ARR-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ARRconcentration in each sample is interpolated from this standard curve.

NCBI Summary for COL4A1    This gene encodes the major type IV alpha collagen chain of basement membranes. Like the other members of the type IV collagen gene family, this gene is organized in a head-to-head conformation with another type IV collagen gene so that each gene pair shares a common promoter. [provided by RefSeq, Jul 2008]

UniProt Comments for COL4A1    COL4A1: Type IV collagen is the major structural component of glomerular basement membranes (GBM), forming a 'chicken-wire' meshwork together with laminins, proteoglycans and entactin/nidogen. Defects in COL4A1 are a cause of brain small vessel disease with hemorrhage (BSVDH). Brain small vessel diseases underlie 20 to 30 percent of ischemic strokes and a larger proportion of intracerebral hemorrhages. Inheritance is autosomal dominant. Defects in COL4A1 are the cause of hereditary angiopathy with nephropathy aneurysms and muscle cramps (HANAC). The clinical renal manifestations include hematuria and bilateral large cysts. Histologic analysis revealed complex basement membrane defects in kidney and skin. The systemic angiopathy appears to affect both small vessels and large arteries. Defects in COL4A1 are a cause of familial porencephaly (POREN1). Porencephaly is a term used for any cavitation or cerebrospinal fluid-filled cyst in the brain. Porencephaly type 1 is usually unilateral and results from focal destructive lesions such as fetal vascular occlusion or birth trauma. Type 2, or schizencephalic porencephaly, is usually symmetric and represents a primary defect or arrest in the development of the cerebral ventricles. Belongs to the type IV collagen family. 2 isoforms of the human protein are produced by alternative splicing.
