 Samples    Serum, plasma, tissue homogenates
 Assay Type    Sandwich
 Detection Range    25 pg/ml -1600 pg/ml.
Sensitivity    The minimum detectable dose of mouse LIPA is typically less than 6.25 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
Intra-assay Precision    Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision    Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.

Preparation and Storage    Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
ISO Certification    Manufactured in an ISO 9001:2008 Certified Laboratory.
Other Notes    Small volumes of LIPA elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for LIPA purchase    MBS922673 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the lipase A, lysosomal acid, cholesterol esterase, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 25 pg/ml -1600 pg/ml in biological research samples containing LIPA, with an estimated sensitivity of The minimum detectable dose of mouse LIPA is typically less than 6.25 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations. The ELISA analytical biochemical technique of the MBS922673 kit is based on LIPA antibody-LIPA antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect LIPA antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, LIPA. Appropriate sample types may include undiluted human body fluids and/or tissue homogenates, secretions such as Serum, plasma, tissue homogenates. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for LIPA elisa kit

Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for LIPA has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any LIPA present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LIPA is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LIPA bound in the initial step. The color development is stopped and the intensity of the color is measured.

UniProt Comments for LIPA    LIPA: Crucial for the intracellular hydrolysis of cholesteryl esters and triglycerides that have been internalized via receptor- mediated endocytosis of lipoprotein particles. Important in mediating the effect of LDL (low density lipoprotein) uptake on suppression of hydroxymethylglutaryl-CoA reductase and activation of endogenous cellular cholesteryl ester formation. Defects in LIPA are the cause of Wolman disease (WOD). WOD is a severe manifestation of LIPA deficiency, leading to the accumulation of cholesteryl esters and triglycerides in most tissues of the body. WOD occurs in infancy and is nearly always fatal before the age of 1 year. Defects in LIPA are the cause of cholesteryl ester storage disease (CESD). CESD is a mild manifestation of LIPA deficiency, leading to the accumulation of cholesteryl esters and triglycerides in most tissues of the body. It is characterized by late-onset. Belongs to the AB hydrolase superfamily. Lipase family. 2 isoforms of the human protein are produced by alternative splicing.
