Specificity    This assay has high sensitivity and excellent specificity for detection of mouse MMP-8. No significant cross-reactivity or interference between mouse MMP-8 and analogues was observed.
Section Bar
Samples    Serum, plasma, cell culture supernates, tissue homogenates
 Assay Type    Sandwich
 Detection Range    15.6 pg/ml -1000 pg/ml.
Sensitivity    The minimum detectable dose of mouse MMP-8 is typically less than 3.9 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
Intra-assay Precision    Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay    Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.

Preparation and Storage    Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
ISO Certification    Manufactured in an ISO 9001:2008 Certified Laboratory.
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of MMP8 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for MMP8 purchase    MBS703729 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the matrix metallopeptidase 8 (neutrophil collagenase) (MMP8) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 15.6 pg/ml -1000 pg/ml in biological research samples containing MMP8, with an estimated sensitivity of The minimum detectable dose of mouse MMP-8 is typically less than 3.9 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations. The ELISA analytical biochemical technique of the MBS703729 kit is based on MMP8 antibody-MMP8 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MMP8 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, MMP8. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernates, tissue homogenates. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%). Mouse matrix metalloproteinase 8, also termed matrix metallopeptidase 8 MMP 8, is unusual in the family of collagenase proteins because it is sequestered in an active form and secreted by neutrophils into serum or plasma. Quantitative detection of mouse free serum plasma or cell culture supernatant neutrophil collagenase MMP8 is possible down to 3.9 pg ml with a sandwich format ELISA kit. The assay system Promega devised utilizes a pre bound polyclonal antibody, biotinylated secondary antibodies and flexible detection products for HRP, either colorimetric or Alexa 174 fluor. Recombinant matrix metalloproteinase 8 protein is included in the ELISA kits for standard curve on each plate, increasing assay precision for MMP8 neutrophil collagenase. Mouse assay kit is specific and does not cross react with MMP8 human or rat. Available in 48 tests or 96 tests formats to order for research purposes such as cell proliferation responses to metallopeptidase 8 neutrophil concentration.

Related Product Information for MMP8 elisa kit

Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MMP-8 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MMP-8 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MMP-8 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MMP-8 bound in the initial step. The color development is stopped and the intensity of the color is measured.

UniProt Comments for MMP8    Function: Can degrade fibrillar type I, II, and III collagens. May play a role in the degradation of collagen fibers during uterine involution.

Catalytic activity: Cleavage of interstitial collagens in the triple helical domain. Unlike EC 3.4.24.7, this enzyme cleaves type III collagen more slowly than type I.

Cofactor: Binds 3 calcium ions per subunit

Enzyme regulation: Cannot be activated without removal of the activation peptide. Activated by matrilysin.
