Searchable Terms for PAPPA purchase    MBS700979 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the pregnancy-associated plasma protein A, pappalysin 1, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 2.5 ng/ml-1000 ng/ml in biological research samples containing PAPPA, with an estimated sensitivity of 2.5 ng/ml. The ELISA analytical biochemical technique of the MBS700979 kit is based on PAPPA antibody-PAPPA antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PAPPA antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PAPPA. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, tissue homogenates. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for PAPPA elisa kit

Introduction: Pregnancy-associated plasma protein A, pappalysin 1, also known as PAPP-A, is a protein used in screening tests for Down syndrome. This protein is a secreted metalloproteinase which cleaves insulin-like growth factor binding proteins (IGFBPs). It is thought to be involved in local proliferative processes such as wound healing and bone remodeling. Low plasma level of this protein has been suggested as a biochemical marker for pregnancies with aneuploid fetuses (fetuses with an abnormal number of chromosomes). For example, low PAPPA may be seen in prenatal screening for Down syndrome.

Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. An antibody specific for PAPP-A antibody has been pre-coated onto a microplate. A competitive inhibition reaction is launched between HRP labeled PAPP-A and unlabeled PAPP-A (Standards or samples) with the antibody specific for PAPP-A. The substrate solutions are added to the wells, 3 respectively. And the color develops in opposite to the amount of PAPP-A bound in the initial step. The color development is stopped and the intensity of the color is measured.

UniProt Comments for PAPPA    Function: Metalloproteinase which specifically cleaves IGFBP-4 and IGFBP-5, resulting in release of bound IGF. Cleavage of IGFBP-4 is dramatically enhanced by the presence of IGF, whereas cleavage of IGFBP-5 is slightly inhibited by the presence of IGF. Isoform 2 cleaves IGFBP-4 very slowly compared to PAPP-A, but its ability to cleave IGFBP-5 is unaffected.1 PublicationManual assertion based on experiment in:Ref.2

Catalytic activity: Cleavage of the 135-Met-|-Lys-136 bond in insulin-like growth factor binding protein (IGFBP)-4, and the 143-Ser-|-Lys-144 bond in IGFBP-5.

Cofactor: Binds 1 zinc ion per subunit
