Specificity    This assay has high sensitivity and excellent specificity for detection of POMC. No significant cross-reactivity or interference between POMC and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete thecross-reactivity detection between POMC and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of POMC elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for POMC purchase    MBS731846 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the pro-opiomelanocortin, POMC, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing POMC, with an estimated sensitivity of 0.1 ng/mL. The ELISA analytical biochemical technique of the MBS731846 kit is based on POMC antibody-POMC antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect POMC antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, POMC. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for POMC elisa kit

Principle of the assay: POMC ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-POMC antibody and an POMC-HRP conjugate. The assay sample and buffer are incubated together with POMC-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the POMC concentration since POMC from samples and POMC-HRP conjugate compete for the anti-POMC antibody binding site. Since the number of sites is limited, as more sites are occupied by POMC from the sample, fewer sites are left to bind POMC-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The POMC concentration in each sample is interpolated from this standard curve.

NCBI Summary for POMC    This gene encodes a polypeptide hormone precursor that undergoes extensive, tissue-specific, post-translational processing via cleavage by subtilisin-like enzymes known as prohormone convertases. There are eight potential cleavage sites within the polypeptide precursor and, depending on tissue type and the available convertases, processing may yield as many as ten biologically active peptides involved in diverse cellular functions. The encoded protein is synthesized mainly in corticotroph cells of the anterior pituitary where four cleavage sites are used; adrenocorticotrophin, essential for normal steroidogenesis and the maintenance of normal adrenal weight, and lipotropin beta are the major end products. In other tissues, including the hypothalamus, placenta, and epithelium, all cleavage sites may be used, giving rise to peptides with roles in pain and energy homeostasis, melanocyte stimulation, and immune modulation. These include several distinct melanotropins, lipotropins, and endorphins that are contained within the adrenocorticotrophin and beta-lipotropin peptides. Mutations in this gene have been associated with early onset obesity, adrenal insufficiency, and red hair pigmentation. Alternatively spliced transcript variants encoding the same protein have been described. [provided by RefSeq]

UniProt Comments for POMC    POMC: ACTH stimulates the adrenal glands to release cortisol. Defects in POMC may be associated with susceptibility to obesity (OBESITY). It is a condition characterized by an increase of body weight beyond the limitation of skeletal and physical requirements, as the result of excessive accumulation of body fat. Defects in POMC are the cause of pro-opiomelanocortinin deficiency (POMCD). Affected individuals present early-onset obesity, adrenal insufficiency and red hair. Belongs to the POMC family.
