Product Description specifically for UBA1 elisa kit

   Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for UBA1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any UBA1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for UBA1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of UBA1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

 NCBI Summary for UBA1    This gene encodes a member of the ubiquitin-activating E1 family. The encoded protein initiates the ubiquitin activation and transfer cascade, catalyzing the first step in ubiquitin conjugation to mark cellular proteins for proteasome degradation. Ubiquitin activating enzymes use ATP to form a thioester between a conserved catalytic cysteine of the enzyme and the C-terminal carboxylate of ubiquitin. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jan 2013]

 UniProt Comments for UBA1    UBE1: an enzyme of the ubiquitin-activating E1 family. Adenylates ubiquitin at its carboxy-terminal glycine residue and thereafter links this residue to the side chain of a cysteine residue in E1, yielding an ubiquitin-E1 thiolester. There are two active sites within the E1 molecule, allowing it to accomodate two ubiquitin moieties at a time, with a new ubiquitin forming an adenylate intermediate as the previous one is transferred to the thiol site. Complements an X-linked mouse temperature-sensitive defect in DNA synthesis, and thus may function in DNA repair.
