Specificity    This assay has high sensitivity and excellent specificity for detection of GAP43. No significant cross-reactivity or interference between GAP43 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between GAP43 and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of GAP43 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for GAP43 purchase    MBS739175 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Growth Associated Protein 43 (GAP43) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing GAP43, with an estimated sensitivity of 0.1ng/mL. The ELISA analytical biochemical technique of the MBS739175 kit is based on GAP43 antibody-GAP43 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect GAP43 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, GAP43. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for GAP43 elisa kit

Intended Uses: This GAP43 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine GAP43. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: GAP43 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GAP43 antibody and an GAP43-HRP conjugate. The assay sample and buffer are incubated together with GAP43-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GAP43 concentration since GAP43 from samples and GAP43-HRP conjugate compete for the anti-GAP43 antibody binding site. Since the number of sites is limited, as more sites are occupied by GAP43 from the sample, fewer sites are left to bind GAP43-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GAP43 concentration in each sample is interpolated from this standard curve.

NCBI Summary for GAP43    The protein encoded by this gene has been termed a 'growth' or 'plasticity' protein because it is expressed at high levels in neuronal growth cones during development and axonal regeneration. This protein is considered a crucial component of an effective regenerative response in the nervous system. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

UniProt Comments for GAP43    GAP43: This protein is associated with nerve growth. It is a major component of the motile growth cones that form the tips of elongating axons. Plays a role in axonal and dendritic filopodia induction. Identified in a complex containing FGFR4, NCAM1, CDH2, PLCG1, FRS2, SRC, SHC1, GAP43 and CTTN. Binds calmodulin with a greater affinity in the absence of Ca(2+) than in its presence. Belongs to the neuromodulin family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Cell surface

Chromosomal Location of Human Ortholog: 3q13.31

Cellular Component: filopodium membrane; axon; cytoplasm; postsynaptic density; plasma membrane; cell junction

Molecular Function: calmodulin binding; protein binding

Biological Process: glial cell differentiation; cell fate commitment; tissue regeneration; regulation of growth; axon choice point recognition; protein kinase C activation; response to wounding; regulation of filopodium formation
