Specificity    This assay has high sensitivity and excellent specificity for detection of LCN-2. No significant cross-reactivity or interference between LCN-2 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between LCN-2 and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of LCN2 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for LCN2 purchase    MBS751396 is a ready-to-use microwell, strip plate Quantitative Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Lipocalin 2 (LCN2) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing LCN2, with an estimated sensitivity of 1.0 ng/mL. The ELISA analytical biochemical technique of the MBS751396 kit is based on LCN2 antibody-LCN2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect LCN2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, LCN2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for LCN2 elisa kit

Intended Uses: This LCN-2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine LCN-2. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay: LCN-2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-LCN-2 antibody and an LCN-2-HRP conjugate. The assay sample and buffer are incubated together with LCN-2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the LCN-2 concentration since LCN-2 from samples and LCN-2-HRP conjugate compete for the anti-LCN-2 antibody binding site. Since the number of sites is limited, as more sites are occupied by LCN-2 from the sample, fewer sites are left to bind LCN-2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The LCN-2 concentration in each sample is interpolated from this standard curve.

UniProt Comments for LCN2    LCN2: Iron-trafficking protein involved in multiple processes such as apoptosis, innate immunity and renal development. Binds iron through association with 2,5-dihydroxybenzoic acid (2,5- DHBA), a siderophore that shares structural similarities with bacterial enterobactin, and delivers or removes iron from the cell, depending on the context. Iron-bound form (holo-24p3) is internalized following binding to the SLC22A17 (24p3R) receptor, leading to release of iron and subsequent increase of intracellular iron concentration. In contrast, association of the iron-free form (apo-24p3) with the SLC22A17 (24p3R) receptor is followed by association with an intracellular siderophore, iron chelation and iron transfer to the extracellular medium, thereby reducing intracellular iron concentration. Involved in apoptosis due to interleukin-3 (IL3) deprivation: iron-loaded form increases intracellular iron concentration without promoting apoptosis, while iron-free form decreases intracellular iron levels, inducing expression of the proapoptotic protein BCL2L11/BIM, resulting in apoptosis. Involved in innate immunity, possibly by sequestrating iron, leading to limit bacterial growth. Belongs to the calycin superfamily. Lipocalin family. 2 isoforms of the human protein are produced by alternative splicing.
