Intended Uses: This S100A12 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine S100A12. This ELISA kit for research use only, not for therapeutic or diagnostic applications!
Principle of the Assay||S100A12 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-S100A12 antibody and an S100A12-HRP conjugate. The assay sample and buffer are incubated together with S100A12-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the S100A12 concentration since S100A12 from samples and S100A12-HRP conjugate compete for the anti-S100A12 antibody binding site. Since the number of sites is limited, as more sites are occupied by S100A12 from the sample, fewer sites are left to bind S100A12-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The S100A12 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. S100 genes include at least 13 members which are located as a cluster on chromosome 1q21. This protein is proposed to be involved in specific calcium-dependent signal transduction pathways and its regulatory effect on cytoskeletal components may modulate various neutrophil activities. Its proinflammatory activity involves recruitment of leukocytes, promotion of cytokine and chemokine production, and regulation of leukocyte adhesion and migration. Acts as an alarmin or a danger associated molecular pattern (DAMP) molecule and stimulates innate immune cells via binding to receptor for advanced glycation endproducts (AGER). Binding to AGER activates the MAP-kinase and NF-kappa-B signaling pathways leading to production of proinflammatory cytokines and up-regulation of cell adhesion molecules ICAM1 and VCAM1.
