Specificity    This assay has high sensitivity and excellent specificity for detection of ACTHR. No significant cross-reactivity or interference between ACTHR and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between ACTHR and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of ACTHR elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for ACTHR purchase    MBS720160 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Adrenocorticotropic Hormone receptor (ACTHR) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 250-5000pg/mL in biological research samples containing ACTHR, with an estimated sensitivity of 1.0pg/mL. The ELISA analytical biochemical technique of the MBS720160 kit is based on ACTHR antibody-ACTHR antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ACTHR antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ACTHR. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for ACTHR elisa kit

Principle of the assay: ACTHR ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ACTHR antibody and an ACTHR-HRP conjugate. The assay sample and buffer are incubated together with ACTHR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACTHR concentration since ACTHR from samples and ACTHR-HRP conjugate compete for the anti-ACTHR antibody binding site. Since the number of sites is limited, as more sites are occupied by ACTHR from the sample, fewer sites are left to bind ACTHR-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACTHR concentration in each sample is interpolated from this standard curve.

NCBI Summary for ACTHR    MC2R encodes one member of the five-member G-protein associated melanocortin receptor family. Melanocortins (melanocyte-stimulating hormones and adrenocorticotropic hormone) are peptides derived from pro-opiomelanocortin (POMC). MC2R is selectively activated by adrenocorticotropic hormone, whereas the other four melanocortin receptors recognize a variety of melanocortin ligands. Mutations in MC2R can result in familial glucocorticoid deficiency. Alternate transcript variants have been found for this gene. [provided by RefSeq, May 2014]
Section Bar
UniProt Comments for ACTHR    MC2R: Receptor for ACTH. This receptor is mediated by G proteins (G(s)) which activate adenylate cyclase. Defects in MC2R are the cause of glucocorticoid deficiency type 1 (GCCD1); also known as familial glucocorticoid deficiency type 1 (FGD1). GCCD1 is an autosomal recessive disorder due to congenital insensitivity or resistance to adrenocorticotropin (ACTH). It is characterized by progressive primary adrenal insufficiency, without mineralocorticoid deficiency. Belongs to the G-protein coupled receptor 1 family.

Protein type: Receptor, GPCR; GPCR, family 1; Membrane protein, integral; Membrane protein, multi-pass

Chromosomal Location of Human Ortholog: 18p11.2

Cellular Component: integral to plasma membrane; cytoplasm; plasma membrane

Molecular Function: protein binding; melanocortin receptor activity; adrenocorticotropin receptor activity

Biological Process: G-protein signaling, coupled to cyclic nucleotide second messenger; G-protein coupled receptor protein signaling pathway; neuropeptide signaling pathway; positive regulation of cAMP biosynthetic process; placenta development

Disease: Glucocorticoid Deficiency 1
