Specificity    This assay has high sensitivity and excellent specificity for detection of MIP-3alpha. No significant cross-reactivity or interference between MIP-3alpha and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between MIP-3alpha and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of MIP-3alpha elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for MIP-3alpha purchase    MBS721691 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Macrophage inflammatory protein-3alpha (MIP-3alpha) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 25-500pg/mL in biological research samples containing MIP-3alpha, with an estimated sensitivity of 1.0pg/mL. The ELISA analytical biochemical technique of the MBS721691 kit is based on MIP-3alpha antibody-MIP-3alpha antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MIP-3alpha antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, MIP-3alpha. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for MIP-3alpha elisa kit

Intended Uses: This MIP-3alpha ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rabbit MIP-3alpha. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay||MIP-3alpha ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MIP-3alpha antibody and an MIP-3alpha-HRP conjugate. The assay sample and buffer are incubated together with MIP-3alpha-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MIP-3alpha concentration since MIP-3alpha from samples and MIP-3alpha-HRP conjugate compete for the anti-MIP-3alpha antibody binding site. Since the number of sites is limited, as more sites are occupied by MIP-3alpha from the sample, fewer sites are left to bind MIP-3alpha-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MIP-3alpha concentration in each sample is interpolated from this standard curve.

UniProt Comments for MIP-3alpha    CCL20: Chemotactic factor that attracts lymphocytes and, slightly, neutrophils, but not monocytes. Inhibits proliferation of myeloid progenitors in colony formation assays. May be involved in formation and function of the mucosal lymphoid tissues by attracting lymphocytes and dendritic cells towards epithelial cells. C-terminal processed forms have been shown to be equally chemotactically active for leukocytes. Possesses antibacterial activity E.coli ATCC 25922 and S.aureus ATCC 29213. Belongs to the intercrine beta (chemokine CC) family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Motility/polarity/chemotaxis; Secreted; Chemokine; Secreted, signal peptide

Cellular Component: extracellular space; extracellular region

Molecular Function: protein binding; chemokine activity; cytokine activity

Biological Process: immune response; chemotaxis; inflammatory response; positive regulation of interleukin-1 alpha biosynthetic process; positive regulation of nitric-oxide synthase biosynthetic process
