Specificity    This assay has high sensitivity and excellent specificity for detection of SPC. No significant cross-reactivity or interference between SPC and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between SPC and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of SPC elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for SPC purchase    MBS724694 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Pulmonary Surfactant Associated Protein C (SPC) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 2.5-50ng/mL in biological research samples containing SPC, with an estimated sensitivity of 0.1ng/mL. The ELISA analytical biochemical technique of the MBS724694 kit is based on SPC antibody-SPC antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect SPC antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, SPC. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for SPC elisa kit

Principle of the assay: SPC ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SPC antibody and an SPC-HRP conjugate. The assay sample and buffer are incubated together with SPC-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SPC concentration since SPC from samples and SPC-HRP conjugate compete for the anti-SPC antibody binding site. Since the number of sites is limited, as more sites are occupied by SPC from the sample, fewer sites are left to bind SPC-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SPC concentration in each sample is interpolated from this standard curve.

UniProt Comments for SPC    SFTPC: Pulmonary surfactant associated proteins promote alveolar stability by lowering the surface tension at the air- liquid interface in the peripheral air spaces. Defects in SFTPC are the cause of pulmonary surfactant metabolism dysfunction type 2 (SMDP2); also called pulmonary alveolar proteinosis due to surfactant protein C deficiency. A rare disease associated with progressive respiratory insufficiency and lung disease with a variable clinical course, due to impaired surfactant homeostasis. It is characterized by alveolar filling with floccular material that stains positive using the periodic acid-Schiff method and is derived from surfactant phospholipids and protein components. Excessive lipoproteins accumulation in the alveoli results in severe respiratory distress. Genetic variations in SFTPC are a cause of susceptibility to respiratory distress syndrome in premature infants (RDS); also known as RDS in prematurity. RDS is a lung disease affecting usually premature newborn infants. It is characterized by deficient gas exchange, diffuse atelectasis, high-permeability lung edema and fibrin-rich alveolar deposits called 'hyaline membranes'. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Lipid-binding
