The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 15.63-1000 pg/mL in biological research samples containing TNF-alpha, with an estimated sensitivity of 9.38 pg/mL. The ELISA analytical biochemical technique of the MBS2500169 kit is based on TNF-alpha antibody-TNF-alpha antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TNF-alpha antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TNF-alpha. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma and other biological fluids. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

   Intended Uses: This ELISA kit can be applied to the in vitro quantitative determination of Rabbit TNF-alpha concentrations in serum, plasma and other biological fluids.

Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been precoated with an antibody specific to Rabbit TNF-alpha. Standards or samples are added to appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibodies specific for Rabbit TNF-alpha and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well successively and incubated. After incubation, free components are washed away. Then the Substrate Reagent is added to each well, only those wells that contain Rabbit TNF-alpha, biotinylated detection antibody and Avidin- HRP conjugate will appear blue in color. The enzyme-substrate reaction will be terminated by adding Stop Solution and appears yellow in color. The optical density (OD) can be measured with spectrophotometry at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of Rabbit TNF-alpha. The concentration of Rabbit TNF-alpha in samples can be calculated by comparing the OD of the samples with the standard curve.

 NCBI Summary for TNF-alpha    This gene encodes a multifunctional proinflammatory cytokine that belongs to the tumor necrosis factor (TNF) superfamily. This cytokine is mainly secreted by macrophages. It can bind to, and thus functions through its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. This cytokine is involved in the regulation of a wide spectrum of biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. This cytokine has been implicated in a variety of diseases, including autoimmune diseases, insulin resistance, and cancer.
