Preparation and Storage    The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
ISO Certification    Manufactured in an ISO 9001:2015 and ISO 13485:2016 Certified Laboratory.
Supply Chain Verification    Manufactured in a lab with traceable raw materials manufactured on site. Coordinated product portfolio of antibodies, pairs, conjugates, recombinant proteins, and immunoassay materials available. Please inquire.
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of ALDOS elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for ALDOS purchase    MBS2022150 is a ready-to-use microwell, strip plate Double-antibody Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Aldosterone Synthase (ALDOS) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 0.156-10ng/mL in biological research samples containing ALDOS, with an estimated sensitivity of < 0.057ng/mL. The ELISA analytical biochemical technique of the MBS2022150 kit is based on ALDOS antibody-ALDOS antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ALDOS antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ALDOS. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, Plasma, Tissue homogenates and Other Biological Fluids. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for ALDOS elisa kit

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Aldosterone Synthase (ALDOS). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Aldosterone Synthase (ALDOS). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Aldosterone Synthase (ALDOS), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Aldosterone Synthase (ALDOS) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

UniProt Comments for ALDOS    CYP11B2: Preferentially catalyzes the conversion of 11- deoxycorticosterone to aldosterone via corticosterone and 18- hydroxycorticosterone. Defects in CYP11B2 are the cause of corticosterone methyloxidase type 1 deficiency (CMO-1 deficiency); also known as aldosterone deficiency due to defect in 18- hydroxylase or aldosterone deficiency I. CMO-1 deficiency is an autosomal recessive disorder of aldosterone biosynthesis. There are two biochemically different forms of selective aldosterone deficiency be termed corticosterone methyloxidase (CMO) deficiency type 1 and type 2. In CMO-1 deficiency, aldosterone is undetectable in plasma, while its immediate precursor, 18- hydroxycorticosterone, is low or normal. Defects in CYP11B2 are the cause of corticosterone methyloxidase type 2 deficiency (CMO-2 deficiency). CMO-2 is an autosomal recessive disorder of aldosterone biosynthesis. In CMO-2 deficiency, aldosterone can be low or normal, but at the expense of increased secretion of 18- hydroxycorticosterone. Consequently, patients have a greatly increased ratio of 18-hydroxycorticosterone to aldosterone and a low ratio of corticosterone to 18-hydroxycorticosterone in serum. Defects in CYP11B2 are a cause of familial hyperaldosteronism type 1 (FH1). It is a disorder characterized by hypertension, variable hyperaldosteronism, and abnormal adrenal steroid production, including 18-oxocortisol and 18-hydroxycortisol. There is significant phenotypic heterogeneity, and some individuals never develop hypertension. The molecular defect causing hyperaldosteronism familial type 1 is an anti-Lepore-type fusion of the CYP11B1 and CYP11B2 genes. The hybrid gene has the promoting part of CYP11B1, ACTH-sensitive, and the coding part of CYP11B2. Belongs to the cytochrome P450 family.

Research Articles on ALDOS    1. Modest increase in aldosterone synthase (AS) expression makes blood pressure more sensitive to salt in mice, suggesting that genetically increased AS expression in humans may contribute to hypertension and cardiovascular complication with high-salt diets.
