Specificity    This assay has high sensitivity and excellent specificity for detection of Rat CASP14. No significant cross-reactivity or interference between Rat CASP14 and analogues was observed. Note: Limited by current skills and knowledge, it is impossible for us to complete the cross- reactivity detection between Rat CASP14 and all the analogues, therefore, cross reaction may still exist.

Searchable Terms for CASP14 purchase    MBS2501126 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the CASP14, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 0.313-20ng/mL in biological research samples containing CASP14, with an estimated sensitivity of Min: 0.188ng/mL; Max: 20ng/mL. The ELISA analytical biochemical technique of the MBS2501126 kit is based on CASP14 antibody-CASP14 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CASP14 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CASP14. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, Plasma, Biological Fluids. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for CASP14 elisa kit

Description: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of CASP14 in Rat serum, plasma and other biological fluids
Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Rat CASP14. Standards or samples are added to the appropriate micro ELISA plate wells and bound by the specific antibody. Then a biotinylated detection antibody specific for Rat CASP14 and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Rat CASP14, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of Rat CASP14. You can calculate the concentration of Rat CASP14 in the samples by comparing the OD of the samples to the standard curve.

NCBI Summary for CASP14    This gene encodes a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. This caspase has been shown to be processed and activated by caspase 8 and caspase 10 in vitro, and by anti-Fas agonist antibody or TNF-related apoptosis inducing ligand in vivo. The expression and processing of this caspase may be involved in keratinocyte terminal differentiation, which is important for the formation of the skin barrier. [provided by RefSeq, Jul 2008]

UniProt Comments for CASP14    CASP14: Believed to be a non-apoptotic caspase which is involved in epidermal differentiation. Seems to play a role in keratinocyte differentiation and cornification. Probably regulates maturation of the epidermis by proteolytically processing filaggrin. Complex of unprocessed caspase-14 and processed 19 kDa (p19) and 10 kDa (p10) subunits. In undifferentiated keratinocytes under postconfluency growth conditions (in vitro). Expressed in keratinocytes of adult skin suprabasal layers (from spinous layers to the stratum granulosum and stratum corneum). Expressed in keratinocytes of hair shaft and sebaceous glands. In psoriatic skin only expressed at very low levels. Belongs to the peptidase C14A family.

Protein type: Protease; EC 3.4.22.-

Chromosomal Location of Human Ortholog: 19p13.1

Cellular Component: keratin filament; cytoplasm; nucleus

Molecular Function: cysteine-type endopeptidase activity

Biological Process: epidermis development; DNA damage response, signal transduction resulting in induction of apoptosis; keratinization; proteolysis

Research Articles on CASP14    1. Mesotrypsin generated saposins A-D from prosaposin, and mature caspase-14 contributed to this process by activating mesotrypsinogen to mesotrypsin. Knockdown of these proteases markedly down-regulated saposin A synthesis in skin equivalent models.
