The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 20 ng/ml-0.312 ng/ml in biological research samples containing CASP9, with an estimated sensitivity of 0.06 ng/ml. The ELISA analytical biochemical technique of the MBS264306 kit is based on CASP9 antibody-CASP9 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CASP9 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CASP9. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Rat serum, plasma or Cell Culture Supernatant and organizations in the natural and recombinant Caspase-9 concentration.    Principle of the assay: This experiment use double-sandwich elisa technique and the ELISA Kit provided is typical. The pre-coated antibody is Rat Caspase-9 monoclonal antibody and the detecting antibody is polyclonal antibody with biotin labeled. Samples and biotin labeling antibody are added into ELISA plate wells and washed out with PBS or TBS. Then Avidin-peroxidase conjugates are added to ELISA wells in order; Use TMB substrate for coloring after reactant thoroughly washed out by PBS or TBS. TMB turns into blue in peroxidase catalytic and finally turns into yellow under the action of acid. The color depth and the testing factors in samples are positively correlated.

 NCBI Summary for CASP9    This gene encodes a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. This protein can undergo autoproteolytic processing and activation by the apoptosome, a protein complex of cytochrome c and the apoptotic peptidase activating factor 1; this step is thought to be one of the earliest in the caspase activation cascade. This protein is thought to play a central role in apoptosis and to be a tumor suppressor. Alternative splicing results in multiple transcript variants. [provided by RefSeq, May 2013]
