The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 62.5-4000pg/mL in biological research samples containing IGFBP7, with an estimated sensitivity of < 26.4pg/mL. The ELISA analytical biochemical technique of the MBS2703574 kit is based on IGFBP7 antibody-IGFBP7 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect IGFBP7 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, IGFBP7. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, Plasma, Tissue Homogenates, Cell Lysates, Cell Culture Supernates and Other Biological Fluids. The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Insulin Like Growth Factor Binding Protein 7 (IGFBP7). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Insulin Like Growth Factor Binding Protein 7 (IGFBP7). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Insulin Like Growth Factor Binding Protein 7 (IGFBP7), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Insulin Like Growth Factor Binding Protein 7 (IGFBP7) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a member of the insulin-like growth factor (IGF)-binding protein (IGFBP) family. IGFBPs bind IGFs with high affinity, and regulate IGF availability in body fluids and tissues and modulate IGF binding to its receptors. This protein binds IGF-I and IGF-II with relatively low affinity, and belongs to a subfamily of low-affinity IGFBPs. It also stimulates prostacyclin production and cell adhesion. Alternatively spliced transcript variants encoding different isoforms have been described for this gene, and one variant has been associated with retinal arterial macroaneurysm (PMID:21835307).
