The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 1.563-100ng/mL in biological research samples containing PKIN1, with an estimated sensitivity of Min: 0.938ng/mL; Max: 100ng/mL. The ELISA analytical biochemical technique of the MBS2512414 kit is based on PKIN1 antibody-PKIN1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PKIN1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PKIN1.    Intended Uses: This ELISA kit applies to the in vitro quantitative determination of Rat PKIN1 concentrations in serum, plasma and other biological fluids.

Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to PKIN1. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for PKIN1 and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain PKIN1, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of PKIN1. You can calculate the concentration of PKIN1 in the samples by comparing the OD of the samples to the standard curve.

 NCBI Summary for PKIN1    The protein encoded by this gene belongs to the protein kinase C superfamily. This kinase is activated by Rho family of small G proteins and may mediate the Rho-dependent signaling pathway. This kinase can be activated by phospholipids and by limited proteolysis. The 3-phosphoinositide dependent protein kinase-1 (PDPK1/PDK1) is reported to phosphorylate this kinase, which may mediate insulin signals to the actin cytoskeleton. The proteolytic activation of this kinase by caspase-3 or related proteases during apoptosis suggests its role in signal transduction related to apoptosis.
