Specificity    This assay has high sensitivity and excellent specificity for detection of SP100B. No significant cross-reactivity or interference between SP100B and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between SP100B and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of S-100B elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for S-100B purchase    MBS731676 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Soluble protein-100B (S-100B) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing S-100B, with an estimated sensitivity of 1.0 pg/mL. The ELISA analytical biochemical technique of the MBS731676 kit is based on S-100B antibody-S-100B antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect S-100B antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, S-100B. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Related Product Information for S-100B elisa kit

Principle of the assay: SP100B ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SP100B antibody and an SP100B-HRP conjugate. The assay sample and buffer are incubated together with SP100B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SP100B concentration since SP100B from samples and SP100B-HRP conjugate compete for the anti-SP100B antibody binding site. Since the number of sites is limited, as more sites are occupied by SP100B from the sample, fewer sites are left to bind SP100B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SP100B concentration in each sample is interpolated from this standard curve.

NCBI Summary for S-100B    The protein encoded by this gene is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. S100 genes include at least 13 members which are located as a cluster on chromosome 1q21; however, this gene is located at 21q22.3. This protein may function in Neurite extension, proliferation of melanoma cells, stimulation of Ca2+ fluxes, inhibition of PKC-mediated phosphorylation, astrocytosis and axonal proliferation, and inhibition of microtubule assembly. Chromosomal rearrangements and altered expression of this gene have been implicated in several neurological, neoplastic, and other types of diseases, including Alzheimer's disease, Down's syndrome, epilepsy, amyotrophic lateral sclerosis, melanoma, and type I diabetes.  UniProt Comments for S-100B    S100B: Weakly binds calcium but binds zinc very tightly- distinct binding sites with different affinities exist for both ions on each monomer. Physiological concentrations of potassium ion antagonize the binding of both divalent cations, especially affecting high-affinity calcium-binding sites. Binds to and initiates the activation of STK38 by releasing autoinhibitory intramolecular interactions within the kinase. Interaction with AGER after myocardial infarction may play a role in myocyte apoptosis by activating ERK1/2 and p53/TP53 signaling. Dimer of either two alpha chains, or two beta chains, or one alpha and one beta chain. The S100B dimer binds two molecules of STK38. Interacts with AGER. The S100B dimer interacts with two molecules of CAPZA1. Although predominant among the water-soluble brain proteins, S100 is also found in a variety of other tissues. Belongs to the S-101 family.
