Specificity    This assay has high sensitivity and excellent specificity for detection of MAFA. No significant cross-reactivity or interference between MAFA and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between MAFA and all the analogues, therefore, cross reaction may still exist in some cases.

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of MAFA elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for MAFA purchase    MBS7210811 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Transcription factor MafA (MAFA) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing MAFA, with an estimated sensitivity of 0.1 ng/mL. The ELISA analytical biochemical technique of the MBS7210811 kit is based on MAFA antibody-MAFA antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect MAFA antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, MAFA. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for MAFA elisa kit

Intended Uses: This MAFA ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat MAFA. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Principle of the Assay||MAFA ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MAFA antibody and an MAFA-HRP conjugate. The assay sample and buffer are incubated together with MAFA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MAFA concentration since MAFA from samples and MAFA-HRP conjugate compete for the anti-MAFA antibody binding site. Since the number of sites is limited, as more sites are occupied by MAFA from the sample, fewer sites are left to bind MAFA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MAFA concentration in each sample is interpolated from this standard curve.

NCBI Summary for MAFA    MAFA is a transcription factor that binds RIPE3b, a conserved enhancer element that regulates pancreatic beta cell-specific expression of the insulin gene (INS; MIM 176730) (Olbrot et al., 2002 [PubMed 12011435]).[supplied by OMIM, Mar 2008]
Section Bar
UniProt Comments for MAFA    MAFA: Acts as a transcriptional factor. Specifically binds the insulin enhancer element RIPE3b and activates insulin gene expression. Cooperates synergistically with NEUROD1 and PDX1. Phosphorylation by GSK3 increases its transcriptional activity and is required for its oncogenic activity. Involved either as an oncogene or as a tumor suppressor, depending on the cell context. Binds DNA as a homodimer (Probable). Interacts with NEUROD1, PCAF and PDX1. Up-regulated by glucose. Belongs to the bZIP family. Maf subfamily.

Protein type: Transcription factor; Tumor suppressor; Oncoprotein; DNA-binding

Chromosomal Location of Human Ortholog: 8q24.3

Cellular Component: nucleus

Molecular Function: protein homodimerization activity; DNA binding; sequence-specific DNA binding; protein heterodimerization activity; transcription factor activity

Biological Process: regulation of transcription, DNA-dependent; transcription, DNA-dependent; insulin secretion; response to glucose stimulus; positive regulation of transcription from RNA polymerase II promoter; nitric oxide mediated signal transduction
