Preparation and Storage    For unopened kit: All the reagents should be kept according to the labels on vials. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C upon receipt while the others should be at 4 degree C.

For opened kit: When the kit is opened, the remaining reagents still need to be stored according to the above storage condition. Besides, please return the unused wells to the foil pouch containing the desiccant pack, and reseal along entire edge of zip-seal.

The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
Product Note    Our ELISA assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay.
Other Notes    Small volumes of TNFRSF10B elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for TNFRSF10B purchase    MBS2025002 is a ready-to-use microwell, strip plate Sandwich ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Tumor Necrosis Factor Receptor Superfamily, Member 10B (TNFRSF10B) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range of 0.156-10ng/mL in biological research samples containing TNFRSF10B, with an estimated sensitivity of Typically less than 0.062ng/mL. The ELISA analytical biochemical technique of the MBS2025002 kit is based on TNFRSF10B antibody-TNFRSF10B antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect TNFRSF10B antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, TNFRSF10B. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Tissue homogenates, Cell lysates and other biological fluids. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for TNFRSF10B elisa kit

Principle of the Assay: The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Tumor Necrosis Factor Receptor Superfamily, Member 10B (TNFRSF10B). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Tumor Necrosis Factor Receptor Superfamily, Member 10B (TNFRSF10B). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Tumor Necrosis Factor Receptor Superfamily, Member 10B (TNFRSF10B), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Tumor Necrosis Factor Receptor Superfamily, Member 10B (TNFRSF10B) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Summary for TNFRSF10B    The protein encoded by this gene is a member of the TNF-receptor superfamily, and contains an intracellular death domain. This receptor can be activated by tumor necrosis factor-related apoptosis inducing ligand (TNFSF10/TRAIL/APO-2L), and transduces an apoptosis signal. Studies with FADD-deficient mice suggested that FADD, a death domain containing adaptor protein, is required for the apoptosis mediated by this protein. Two transcript variants encoding different isoforms and one non-coding transcript have been found for this gene. [provided by RefSeq, Mar 2009]

UniProt Comments for TNFRSF10B    TRAIL-R2: Receptor for the cytotoxic ligand TNFSF10/TRAIL. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. Promotes the activation of NF- kappa-B. Homotrimer. Can interact with TRADD and RIPK1. Regulated by p53/TP53. Widely expressed in adult and fetal tissues; very highly expressed in tumor cell lines such as HeLaS3, K-562, HL-60, SW480, A-549 and G-361; highly expressed in heart, peripheral blood lymphocytes, liver, pancreas, spleen, thymus, prostate, ovary, uterus, placenta, testis, esophagus, stomach and throughout the intestinal tract; not detectable in brain. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, integral; Motility/polarity/chemotaxis

Chromosomal Location of Human Ortholog: 8p22-p21

Cellular Component: plasma membrane; integral to membrane

Molecular Function: protein binding; TRAIL binding; receptor activity

Biological Process: regulation of apoptosis; caspase activation; positive regulation of I-kappaB kinase/NF-kappaB cascade; cell surface receptor linked signal transduction; induction of apoptosis via death domain receptors; apoptosis; activation of NF-kappaB-inducing kinase

Disease: Squamous Cell Carcinoma, Head And Neck
