The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing ADCYAP1. The ELISA analytical biochemical technique of the MBS706315 kit is based on ADCYAP1 antibody-ADCYAP1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect ADCYAP1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, ADCYAP1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

   Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. An antibody specific to ADCYAP1 has been pre-coated onto a microplate. Standards or samples are added to the appropriate microtiter plate wells with biotin-conjugated ADCYAP1 and incubated. A competitive inhibition reaction is launched between ADCYAP1 (Standards or samples) and Biotin-conjugated ADCYAP1 with the pre-coated antibody specific for ADCYAP1. The more amount of ADCYAP1 in samples, the less antibody bound by Biotin-conjugated ADCYAP1. Then Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The substrate solutions are added to the wells, respectively. And the color develops in opposite to the amount of ADCYAP1 in the sample. The color development is stopped and the intensity of the color is measured.
