Specificity    This assay has high sensitivity and excellent specificity for detection of VDBP. No significant cross-reactivity or interference between VDBP and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between VDBP and all the analogues, therefore, cross reaction may still exist in some cases.

Samples    Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate
 Assay Type    Competitive
 Sensitivity    1.0 ng/mL.
Intended Uses    This VDBP ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Sheep VDBP. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

Preparation and Storage    Store all reagents at 2-8 degree C.
Sample Preparation    We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of GC elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for GC purchase    MBS7214240 is a ready-to-use microwell, strip plate Competitive ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the vitamin D-binding protein (GC) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing GC, with an estimated sensitivity of 1.0 ng/mL. The ELISA analytical biochemical technique of the MBS7214240 kit is based on GC antibody-GC antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect GC antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, GC. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Product Description specifically for GC elisa kit

Principle of the assay: VDBP ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-VDBP antibody and an VDBP-HRP conjugate. The assay sample and buffer are incubated together with VDBP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the VDBP concentration since VDBP from samples and VDBP-HRP conjugate compete for the anti-VDBP antibody binding site. Since the number of sites is limited, as more sites are occupied by VDBP from the sample, fewer sites are left to bind VDBP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The VDBP concentration in each sample is interpolated from this standard curve.

NCBI Summary for GC    The protein encoded by this gene belongs to the albumin gene family. It is a multifunctional protein found in plasma, ascitic fluid, cerebrospinal fluid and on the surface of many cell types. It binds to vitamin D and its plasma metabolites and transports them to target tissues. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Feb 2011]

UniProt Comments for GC    GC: Multifunctional protein found in plasma, ascitic fluid, cerebrospinal fluid, and urine and on the surface of many cell types. In plasma, it carries the vitamin D sterols and prevents polymerization of actin by binding its monomers. DBP associates with membrane-bound immunoglobulin on the surface of B-lymphocytes and with IgG Fc receptor on the membranes of T-lymphocytes. Belongs to the ALB/AFP/VDB family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Secreted, signal peptide; Secreted

Chromosomal Location of Human Ortholog: 4q12-q13

Cellular Component: extracellular space; lysosomal lumen; extracellular region; cytosol

Molecular Function: vitamin D binding; actin binding; vitamin transporter activity

Biological Process: steroid metabolic process; vitamin transport; vitamin D metabolic process

Disease: Graves Disease, Susceptibility To, 1

Research Articles on GC    1. GC1s haplotype carriage may increase the risk of RSV bronchiolitis in infancy and subsequent asthma development. The GC1s haplotype is associated with higher VDBP levels, resulting in less freely available vitamin D.
