Samples    Immunoperoxidase Assay for Determination of Vitamin D Binding Protein in Human Samples
Intended Uses    The principle of the double antibody sandwich ELISA is represented in Figure 1. In this assay the VDBP present in samples reacts with the anti-VDBP antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, anti-VDBP antibodies conjugated with horseradish peroxidase (HRP), are added. These enzyme-labeled antibodies form complexes with the previously bound VDBP. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of VDBP in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of VDBP in the test sample. The quantity of VDBP in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

Preparation and Storage    Store at 4 degree C.
Product Note    Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes    Small volumes of GC elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.
Searchable Terms for GC purchase    MBS564062 is a ready-to-use microwell, strip-or-full plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Human Vitamin D Binding Protein (GC) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing GC. The ELISA analytical biochemical technique of the MBS564062 kit is based on GC antibody-GC antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect GC antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, GC. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions such as Immunoperoxidase Assay for Determination of Vitamin D Binding Protein in Human Samples. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

NCBI Summary for GC    The protein encoded by this gene belongs to the albumin gene family. It is a multifunctional protein found in plasma, ascitic fluid, cerebrospinal fluid and on the surface of many cell types. It binds to vitamin D and its plasma metabolites and transports them to target tissues. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq]

UniProt Comments for GC    GC: Multifunctional protein found in plasma, ascitic fluid, cerebrospinal fluid, and urine and on the surface of many cell types. In plasma, it carries the vitamin D sterols and prevents polymerization of actin by binding its monomers. DBP associates with membrane-bound immunoglobulin on the surface of B-lymphocytes and with IgG Fc receptor on the membranes of T-lymphocytes. Belongs to the ALB/AFP/VDB family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Secreted; Secreted, signal peptide

Chromosomal Location of Human Ortholog: 4q12-q13

Cellular Component: extracellular space; lysosomal lumen; extracellular region; cytosol

Molecular Function: vitamin D binding; actin binding; vitamin transporter activity
