 Host Note    Optimized DNA sequence encoding Human extracellular domain of human CD155 including a C-terminal 6His tag was expressed in HEK293 cells.
 Biological Activity    The biological activity of Human PVR was measured by the binding ability of immobilized recombinant Human CD155 (2ug/ml, 100 ul/well) to Human DNAM1 (linear rande of 1-32 ng/ml) in a functional ELISA assay.
 Reconsitution    A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1mg/ml. This solution can then be diluted into other buffers.
 Endotoxin    Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/ug (1EU/ug).
 Molecular Weight Note    Recombinant CD155 is a monomer protein consisting of 329 amino acid residue subunits, due to glycosylation migrates as an approximately 55-60 kDa protein on SDS-PAGE.

 Preparation and Storage    Recombinant CD155, as supplied, can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C to -70 degree C for twelve months.

Avoid repeated freeze/thaw cycles.
 Other Notes    Small volumes of CD155 recombinant protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

 NCBI Summary for CD155    The protein encoded by this gene is a transmembrane glycoprotein belonging to the immunoglobulin superfamily. The external domain mediates cell attachment to the extracellular matrix molecule vitronectin, while its intracellular domain interacts with the dynein light chain Tctex-1/DYNLT1. The gene is specific to the primate lineage, and serves as a cellular receptor for poliovirus in the first step of poliovirus replication.  UniProt Comments for CD155    PVR: Mediates NK cell adhesion and triggers NK cell effector functions. Binds two different NK cell receptors: CD96 and CD226. These interactions accumulates at the cell-cell contact site, leading to the formation of a mature immunological synapse between NK cell and target cell. This may trigger adhesion and secretion of lytic granules and IFN-gamma and activate cytoxicity of activated NK cells. May also promote NK cell-target cell modular exchange, and PVR transfer to the NK cell. This transfer is more important in some tumor cells expressing a lot of PVR, and may trigger fratricide NK cell activation, providing tumors with a mechanism of immunoevasion. Plays a role in mediating tumor cell invasion and migration. Serves as a receptor for poliovirus attachment to target cells. May play a role in axonal transport of poliovirus, by targeting virion-PVR-containing endocytic vesicles to the microtubular network through interaction with DYNLT1. This interaction would drive the virus-containing vesicle to the axonal retrograde transport. Belongs to the nectin family. 4 isoforms of the human protein are produced by alternative splicing.
