Host Note    DNA sequence encoding immunodominant fragment of Cytomegalovirus 65kDa tegument phosphoprotein was expressed in Escherichia Coli.
Reconstitution    A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Molecular Weight Note    Recombinant CMV pp65 (UL83) encodes the immunodominant sequences of the Cytomegalovirus matrix phosphoprotein. This molecule has a calculated mass of approximately 22kDa.

Recombinant CMV pp65 contains an N terminal GST purification tag and migrates as an approximately 52 kDa protein under non-reducing conditions and reducing conditions in SDS-PAGE.

Preparation and Storage    Upon reconstitution, this protein can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.
Other Notes    Small volumes of CMV pp65 recombinant protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

Related Product Information for CMV pp65 recombinant protein

Human cytomegalovirus (HCMV) lower matrix protein pp65 (ppUL83), a major target for host anti-HCMV response, is the product of a late-expressed virus mRNA (reviewed in Spaete et al., 1994). pp65 replenishes a type of HCMV non-infectious particle, the dense bodies, which are secreted by infected cells along with virionsThe phosphoprotein pp65 (ppUL83) of human cytomegalovirus (HCMV) is abundantly synthesized during lytic infection in cultured fibroblasts. As a major constituent of extracellular particles, it gains entry to infected cells immediately after adsorption and subsequently translocates to the cell nucleus.

Application Notes for CMV pp65 recombinant protein    CMV pp65 antigen is suitable for ELISA and Western blots, excellent antigen for detection of CMV with minimal specificity problems.

UniProt Comments for CMV pp65    Function: Counteracts the host antiviral immune response when activated and phosphorylated, by preventing IRF3 from entering the nucleus. Also participates in the transactivation of viral major immediate-early genes by the recruitment of host IFI16 to the promoters pf these genes. Ref.9 Ref.11

Subunit structure: Interacts with host NCL/nucleolin. Interacts with host IFI16. Ref.11 Ref.12

Subcellular location: Virion tegument

Potential. Host nucleus. Host cytoplasm. Note: As part of the incoming virion, pp65 is targeted to the nucleus immediately after infection. The newly synthesized pp65 is observed in the nucleus until some time after 48 hours postinfection. Thereafter, pp65 is probably exported and accumulates in the cytoplasm. Also found in dense bodies. Ref.6 Ref.10

Post-translational modification: Phosphorylation may play a role in the localization of the protein.

Sequence similarities: Belongs to the herpesviridae pp65 family.
