 3D Structure    ModBase 3D Structure for P03172
 Host    E Coli
Section Bar
 Purity/Purification    >95%, as determined by SDS-PAGE.
 Form/Format    Lyophilized from 25mM Tris-Hcl pH 7.2, 1mM EDTA and 50% glycerol.
Section Bar
Host Note    DNA sequence encoding immunodominant fragment of human Herpesvirus-2 Glycoprotein D was expressed in Escherichia Coli.
Reconstitution    A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Molecular Weight Note    Recombinant HSV-2 gD encodes the immunodominant sequences of the human Herpesvirus Glycoprotein D. This molecule has a calculated mass of approximately 14 kDa.

Recombinant HSV2 gD contains an N terminal GST purification tag and migrates as an approximately 40 kDa protein under non-reducing conditions and reducing conditions in SDS-PAGE.
Section Bar
Preparation and Storage    The lyophilized protein is stable for at least 2 years from date of receipt at -20 degree C.
Upon reconstitution, this protein can be stored in working aliquots at 2 degree - 8 degree C for one month, or at -20 degree C for six months, with a carrier protein without detectable loss of activity.

Avoid repeated freeze/thaw cycles.
Other Notes    Small volumes of HSV-2 gD recombinant protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

Related Product Information for HSV-2 gD recombinant protein

Herpes simplex virus (HSV) glycoprotein D (gD) is essential for virus entry into cells, as well as for cell fusion and cell-to-cell spread. The functions of gD are best understood for virus entry, a process in which gD binds to "gD receptors," cell surface molecules that are more restricted in number than the glycosaminoglycans to which the virus initially adsorbs.
Product Categories/Family for HSV-2 gD recombinant protein    Viral Antigens
Application Notes for HSV-2 gD recombinant protein    Recombinant human Herpesvirus-2 gD antigen is suitable for ELISA and Western blots, excellent antigen for detection of HSV with minimal specificity problems.

UniProt Comments for HSV-2 gD    Function: Envelope glycoprotein that binds to the potential host cell entry receptors TNFRSF14/HVEM, PVRL1 and PVRL1. May trigger fusion with host membrane, by recruiting the fusion machinery composed of gB and gH/gL

By similarity.

Subunit structure: Homodimer

By similarity. Interacts with host receptor TNFRSF14. Interacts with host receptor PVRL1. Interacts with host receptor PVRL2. Interacts (via profusion domain) with gB; this interaction occurs in the absence of gH/gL. Interacts (via profusion domain) with gH/gL heterodimer; this interaction occurs in the absence of gB. Associates with the gB-gH/gL-gD complex. Interacts (via C-terminus) with UL11 tegument protein

By similarity. Ref.6 Ref.7

Subcellular location: Virion membrane; Single-pass type I membrane protein

By similarity. Note: During virion morphogenesis, this protein probably accumulates in the endosomes and trans-Golgi where secondary envelopment occurs

By similarity.

Sequence similarities: Belongs to the herpesviridae glycoprotein D family.
