Lysozyme lyses the peptidoglycan polymer typically found in most bacterial cell walls. Recombinant Lysozyme has identical amino acid sequences as the native Lysozyme. In lysing bacterial cells, recombinant Lysozyme is added to the cell extraction buffer at a range of 0.05 to 0.5 mg/mL (7,000 to 70,000 U/mL).

Typically, the extraction buffer contains 100mM tris HCL, 2mM EDTA, 0.05% trion I-100, pH 8.0. The cell paste is suspended in extraction buffer and incubated for at least 15 minutes at room temperature. Alternatively, Triton X-100 at 1% and PMSF at 1mM can be added separately and the lysis solution is further incubated for at least 15 minutes at 37C. The lysed cell suspension is clarified by centrifugation at 20,000 g for at least 15 minutes. The extraction conditions and concentration of recombinant lyosozyme needed for optiomal cell lysis and protein recovery depends on various factors.

Not only does lysozyme have very important clinical implications, but it is also a useful tool in molecular biology. The ability of lysozyme to break down bacterial cell walls (cell lysis) is utilized by molecular biologists. Cell lysis is necessary to extract recombinant proteins or DNA from bacterial cells. Human lysozyme is preferred to chicken lysozyme because it is up to four times more active. Also, the activity of chicken lysozyme has high lot-to-lot variation. Many recombinant protein production systems prefer an animal-free lysozyme to avoid the risk of infectious disease transmission.

 NCBI Summary for LYZ    This gene encodes human lysozyme, whose natural substrate is the bacterial cell wall peptidoglycan (cleaving the beta[1-4]glycosidic linkages between N-acetylmuramic acid and N-acetylglucosamine). Lysozyme is one of the antimicrobial agents found in human milk, and is also present in spleen, lung, kidney, white blood cells, plasma, saliva, and tears. The protein has antibacterial activity against a number of bacterial species. Missense mutations in this gene have been identified in heritable renal amyloidosis. [provided by RefSeq, Oct 2014]

 UniProt Comments for LYZ    LYZ: Lysozymes have primarily a bacteriolytic function; those in tissues and body fluids are associated with the monocyte- macrophage system and enhance the activity of immunoagents. Defects in LYZ are a cause of amyloidosis type 8 (AMYL8); also known as systemic non-neuropathic amyloidosis or Ostertag-type amyloidosis. AMYL8 is a hereditary generalized amyloidosis due to deposition of apolipoprotein A1, fibrinogen and lysozyme amyloids. Viscera are particularly affected. There is no involvement of the nervous system. Clinical features include renal amyloidosis resulting in nephrotic syndrome, arterial hypertension, hepatosplenomegaly, cholestasis, petechial skin rash. Belongs to the glycosyl hydrolase 22 family.
