Purity/Purification    Greater than 99.0% as determined by:
(a) Gel filtration analysis.
(b) Analysis by reducing and non-reducing SDS-PAGE gel.
(c) Analysis by RP-HPLC.
 Form/Format    White lyophilized (freeze-dried) powder.
Section Bar
Solubility    It is recommended to reconstitute the lyophilized zfGH in sterile water or 0.4% NaHCO3 adjusted to pH 8-9, not less than 100ug/ml and not more than 3 mg/ml, which can then be further diluted to other aqueous solutions, preferably in presence of carrier protein.
Protein Content    Protein quantitation was carried out by UV spectroscopy at 280 nm using the absorbency value of 0.42 as the extinction coefficient for a 0.1% (1mg/ml) solution at pH 8.0 This value is calculated by the DNAman program.
Dimers and Aggregates    The purified zfGH (21K) consists of > 99% monomers as determined by gel-filtration chromatography.
 Endotoxin    Less than 0.05 ng/ug (IEU/ug) of zfGH (22 K).
Formulation    The protein was lyophilized from a concentrated (1mg/ml) solution with 0.5% NaHCO3 adjusted to pH 8.
Biological Activity    zfGH is biologically active in PDF-P1 3B9 cells stably transfected with rabbit GH receptors.
Section Bar
Preparation and Storage    Lyophilized zfGH although stable at room temperature for 3 weeks, should be stored desiccated below -18 degree C. Upon reconstitution at > 0.1 mg/ml and filter sterilization zfGH can be stored at 4 degree C for several weeks. At lower concentration addition of a carrier protein (0.1% HSA or BSA) is recomended. Please prevent freeze-thaw cycles.
Other Notes    Small volumes of zfGH recombinant protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for zfGH    The protein encoded by this gene is a member of the somatotropin/prolactin family of hormones which play an important role in growth control. The gene, along with four other related genes, is located at the growth hormone locus on chromosome 17 where they are interspersed in the same transcriptional orientation; an arrangement which is thought to have evolved by a series of gene duplications. The five genes share a remarkably high degree of sequence identity. Alternative splicing generates additional isoforms of each of the five growth hormones, leading to further diversity and potential for specialization. This particular family member is expressed in the pituitary but not in placental tissue as is the case for the other four genes in the growth hormone locus. Mutations in or deletions of the gene lead to growth hormone deficiency and short stature. [provided by RefSeq, Jul 2008]

UniProt Comments for zfGH    GH: Plays an important role in growth control. Its major role in stimulating body growth is to stimulate the liver and other tissues to secrete IGF-1. It stimulates both the differentiation and proliferation of myoblasts. It also stimulates amino acid uptake and protein synthesis in muscle and other tissues. Defects in GH1 are a cause of growth hormone deficiency isolated type 1A (IGHD1A); also known as pituitary dwarfism I. IGHD1A is an autosomal recessive deficiency of GH which causes short stature. IGHD1A patients have an absence of GH with severe dwarfism and often develop anti-GH antibodies when given exogenous GH. Defects in GH1 are a cause of growth hormone deficiency isolated type 1B (IGHD1B); also known as dwarfism of Sindh. IGHD1B is an autosomal recessive deficiency of GH which causes short stature. IGHD1B patients have low but detectable levels of GH. Dwarfism is less severe than in IGHD1A and patients usually respond well to exogenous GH. Defects in GH1 are the cause of Kowarski syndrome (KWKS); also known as pituitary dwarfism VI. Defects in GH1 are a cause of growth hormone deficiency isolated type 2 (IGHD2). IGHD2 is an autosomal dominant deficiency of GH which causes short stature. Clinical severity is variable. Patients have a positive response and immunologic tolerance to growth hormone therapy. Belongs to the somatotropin/prolactin family. 4 isoforms of the human protein are produced by alternative splicing.

Research Articles on zfGH    1. Analysis of GH1 in a cohort of Brazilian patients revealed that the autosomal recessive form of isolated growth hormone deficiency(IGHD) was more common than the dominant one, and both were found only in severe IGHD.
