 Quality Control    The sequence of shRNA is guaranteed by sequencing.
 Directions for Use    Resuspend lyophilized shRNA plasmid DNA in 500 ul of the deionized water. Each vial contains 50 ug of lyophilized shRNA plasmid DNA. Suitable for up to 50 transfections.
 Components    We offer 3 different plasmids which knock down human QTRT1 gene specifically. Each vial contains 50 ug of lyophilized shRNA.

 Preparation and Storage    Store lyophilized shRNA plasmid DNA at -20 degree C with desiccant. Stable for one year. Once resuspended, store at 4 degree C for short term storage or -80 degree C for long term storage. Avoid repeated freeze thaw cycles.

 NCBI Summary for QTRT1    This gene encodes the catalytic subunit of tRNA-guanine transglycosylase. tRNA-guanine transglycosylase is a heterodimeric enzyme complex that plays a critical role in tRNA modification by synthesizing the 7-deazaguanosine queuosine, which is found in tRNAs that code for asparagine, aspartic acid, histidine and tyrosine. A pseudogene of this gene is located on the long arm of chromosome X. [provided by RefSeq, Feb 2012]

 UniProt Comments for QTRT1    Catalytic subunit of the queuine tRNA-ribosyltransferase (TGT) that catalyzes the base-exchange of a guanine (G) residue with queuine (Q) at position 34 (anticodon wobble position) in tRNAs with GUN anticodons (tRNA-Asp, -Asn, -His and -Tyr), resulting in the hypermodified nucleoside queuosine (7-(((4,5-cis-dihydroxy-2-cyclopenten-1-yl)amino)methyl)-7-deazaguanosine) (PubMed:11255023, PubMed:20354154). Catalysis occurs through a double-displacement mechanism. The nucleophile active site attacks the C1' of nucleotide 34 to detach the guanine base from the RNA, forming a covalent enzyme-RNA intermediate. The proton acceptor active site deprotonates the incoming queuine, allowing a nucleophilic attack on the C1' of the ribose to form the product ().
