The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing PSG4. The ELISA analytical biochemical technique of the MBS916501 kit is based on PSG4 antibody-PSG4 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PSG4 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PSG4. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the competitive enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with goat-anti-rabbit antibody. Standards or samples are added to the appropriate microtiter plate wells with an antibody specific for PSG4 and Horseradish Peroxidase (HRP) conjugated PSG4. The competitive inhibition reaction is launched between with HRP labeled PSG4 and unlabeled PSG4 with the antibody. A substrate solution is added to the wells and the color develops in opposite to the amount of PSG4 in the sample. The color development is stopped and the intensity of the color is measured.

The human pregnancy-specific glycoproteins (PSGs) are a family of proteins that are synthesized in large amounts by placental trophoblasts and released into the maternal circulation during pregnancy. Molecular cloning and analysis of several PSG genes has indicated that the PSGs form a subgroup of the carcinoembryonic antigen (CEA) gene family, which belongs to the immunoglobulin superfamily of genes. Members of the CEA family consist of a single N domain, with structural similarity to the immunoglobulin variable domains, followed by a variable number of immunoglobulin constant-like A and/or B domains. Most PSGs have an arg-gly-asp (RGD) motif, which has been shown to function as an adhesion recognition signal for several integrins, in the N-terminal domain (summary by Teglund et al., 1994 [PubMed 7851896]).
