The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing PTGDS. The ELISA analytical biochemical technique of the MBS917167 kit is based on PTGDS antibody-PTGDS antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect PTGDS antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, PTGDS. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for PTGDS has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PTGDS present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PTGDS is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PTGDS bound in the initial step. The color development is stopped and the intensity of the color is measured.

Function: Catalyzes the conversion of PGH2 to PGD2, a prostaglandin involved in smooth muscle contraction/relaxation and a potent inhibitor of platelet aggregation. Involved in a variety of CNS functions, such as sedation, NREM sleep and PGE2-induced allodynia, and may have an anti-apoptotic role in oligodendrocytes. Binds small non-substrate lipophilic molecules, including biliverdin, bilirubin, retinal, retinoic acid and thyroid hormone, and may act as a scavenger for harmful hydrophopic molecules and as a secretory retinoid and thyroid hormone transporter. Possibly involved in development and maintenance of the blood-brain, blood-retina, blood-aqueous humor and blood-testis barrier.
