The ELISA analytical biochemical technique of the MBS702772 kit is based on Renin antibody-Renin antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect Renin antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, Renin. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for renin has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any renin present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for renin is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of renin bound in the initial step. The color development is stopped and the intensity of the color is measured.

Renin catalyzes the first step in the activation pathway of angiotensinogen--a cascade that can result in aldosterone release,vasoconstriction, and increase in blood pressure. Renin, an aspartyl protease, cleaves angiotensinogen to form angiotensin I, which is converted to angiotensin II by angiotensin I converting enzyme, an important regulator of blood pressure and electrolyte balance. Transcript variants that encode different protein isoforms and that arise from alternative splicing and the use of alternative promoters have been described, but their full-length nature has not been determined. Mutations in this gene have been shown to cause familial hyperproreninemia. [provided by RefSeq, Jul 2008]
