The ELISA Kit is designed to detect native, not recombinant, RPLP1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Intended Uses: This RPLP1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human RPLP1.

Principle of the Assay: RPLP1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-RPLP1 antibody and an RPLP1-HRP conjugate. The assay sample and buffer are incubated together with RPLP1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RPLP1 concentration since RPLP1 from samples and RPLP1-HRP conjugate compete for the anti-RPLP1 antibody binding site. Since the number of sites is limited, as more sites are occupied by RPLP1 from the sample, fewer sites are left to bind RPLP1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RPLP1 concentration in each sample is interpolated from this standard curve.

Ribosomes, the organelles that catalyze protein synthesis, consist of a small 40S subunit and a large 60S subunit. Together these subunits are composed of 4 RNA species and approximately 80 structurally distinct proteins. This gene encodes a ribosomal phosphoprotein that is a component of the 60S subunit. The protein, which is a functional equivalent of the E. coli L7/L12 ribosomal protein, belongs to the L12P family of ribosomal proteins. It plays an important role in the elongation step of protein synthesis. Unlike most ribosomal proteins, which are basic, the encoded protein is acidic. Its C-terminal end is nearly identical to the C-terminal ends of the ribosomal phosphoproteins P0 and P2. The P1 protein can interact with P0 and P2 to form a pentameric complex consisting of P1 and P2 dimers, and a P0 monomer. The protein is located in the cytoplasm. Two alternatively spliced transcript variants that encode different proteins have been observed.
