MBS178573 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the SAA/SAA1, ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing SAA1. The ELISA analytical biochemical technique of the MBS178573 kit is based on SAA1 antibody-SAA1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect SAA1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, SAA1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: Human SAA/SAA1 ELISA Kit was based on standard sandwich enzyme-linked immunesorbent assay technology. A monoclonal antibody from mouse specific for SAA/SAA1 has been precoated onto 96-well plates. Standards(Expression system for standard: E.coli; Immunogen sequence: S20-Y122) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for SAA/SAA1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human SAA/SAA1 amount of sample captured in plate.

Background/Introduction: Serum amyloid A protein (SAA), also known as SAA1, is a protein that in humans is encoded by the SAA1 gene. This gene encodes a member of the serum amyloid A family of apolipoproteins. It is mapped to 11p15.1. SAA is a major acute phase protein that is highly expressed in response to inflammation and tissue injury. This protein also plays an important role in HDL metabolism and cholesterol homeostasis. High levels of this protein are associated with chronic inflammatory diseases including atherosclerosis, rheumatoid arthritis, Alzheimer's disease and Crohn's disease. This protein may also be a potential biomarker for certain tumors.

NCBI Summary for SAA1
This gene encodes a member of the serum amyloid A family of apolipoproteins. The encoded preproprotein is proteolytically processed to generate the mature protein. This protein is a major acute phase protein that is highly expressed in response to inflammation and tissue injury. Alternate splicing results in multiple transcript variants that encode the same protein. A pseudogene of this gene is found on chromosome 11. [provided by RefSeq, Feb 2016]
UniProt Comments for SAA1
SAA1: Major acute phase reactant. Apolipoprotein of the HDL complex. Reactive, secondary amyloidosis is characterized by the extracellular accumulation in various tissues of the SAA1 protein. These deposits are highly insoluble and resistant to proteolysis; they disrupt tissue structure and compromise function. Elevated serum SAA1 protein levels may be associated with lung cancer. Belongs to the SAA family.

Protein type: Secreted; Secreted, signal peptide

Chromosomal Location of Human Ortholog: 11p15.1

Cellular Component: cytoplasmic microtubule; extracellular region; extracellular space

Molecular Function: chemoattractant activity; G-protein-coupled receptor binding

Biological Process: activation of MAPK activity; cellular protein metabolic process; elevation of cytosolic calcium ion concentration; innate immune response; lymphocyte chemotaxis; macrophage chemotaxis; positive regulation of cell adhesion; positive regulation of cytokine secretion; receptor-mediated endocytosis
