The ELISA analytical biochemical technique of the MBS2000212 kit is based on SAT1 antibody-SAT1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect SAT1 antigen targets in samples. The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Spermidine/Spermine N1-Acetyltransferase 1 (SAT1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Spermidine/Spermine N1-Acetyltransferase 1 (SAT1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Spermidine/Spermine N1-Acetyltransferase 1 (SAT1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Spermidine/Spermine N1-Acetyltransferase 1 (SAT1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

The protein encoded by this gene belongs to the acetyltransferase family, and is a rate-limiting enzyme in the catabolic pathway of polyamine metabolism. It catalyzes the acetylation of spermidine and spermine, and is involved in the regulation of the intracellular concentration of polyamines and their transport out of cells. Defects in this gene are associated with keratosis follicularis spinulosa decalvans (KFSD). Alternatively spliced transcripts have been found for this gene.[provided by RefSeq, Sep 2009]

This highly regulated enzyme allows a fine attenuation of the intracellular concentration of polyamines. Also involved in the regulation of polyamine transport out of cells. Acts on 1,3-diaminopropane, 1,5-diaminopentane, putrescine, spermidine (forming N(1)- and N(8)-acetylspermidine), spermine, N(1)-acetylspermidine and N(8)-acetylspermidine. Defects in SAT1 may be a cause of keratosis follicularis spinulosa decalvans X-linked (KFSDX). A rare disorder affecting the skin and the eye.
