The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing SIRPa. The ELISA analytical biochemical technique of the MBS2019546 kit is based on SIRPa antibody-SIRPa antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect SIRPa antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, SIRPa. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Signal Regulatory Protein Alpha (SIRPa). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Signal Regulatory Protein Alpha (SIRPa). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Signal Regulatory Protein Alpha (SIRPa), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Signal Regulatory Protein Alpha (SIRPa) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

The protein encoded by this gene is a member of the signal-regulatory-protein (SIRP) family, and also belongs to the immunoglobulin superfamily. SIRP family members are receptor-type transmembrane glycoproteins known to be involved in the negative regulation of receptor tyrosine kinase-coupled signaling processes. This protein can be phosphorylated by tyrosine kinases. The phospho-tyrosine residues of this PTP have been shown to recruit SH2 domain containing tyrosine phosphatases (PTP), and serve as substrates of PTPs. This protein was found to participate in signal transduction mediated by various growth factor receptors. CD47 has been demonstrated to be a ligand for this receptor protein. This gene and its product share very high similarity with several other members of the SIRP family. These related genes are located in close proximity to each other on chromosome 20p13. Multiple alternatively spliced transcript variants have been determined for this gene. [provided by RefSeq, Jul 2008]
